With the aid of a few on here, my first app is ready to go. Builds fine, no problem. However, the app icon on my desktop (unzipped) has a grey and white ‘prevent/stop’ circle on it. Does this go when I start the upload to app store process? I don’t want to start the process and then find I have missed something obvious!

No, no issue - that’s what it looks like to all of us. Just like when you load an Adhoc build into iTunes it wont show the actual icon.
On the store it will be fine so long as the icon shows when you load it onto your device (as in, it isn’t just a white icon on your iPhone/iPad/etc).
