I just have access to Kindle Fire and Galaxy Tab for Android Testing. Is it possible to build for Nook and release the app. on Nook store without actually testing on Nook device? Assuming that it works fine on other similar specs. tablets?

It is possible to submit without testing on device, yes - however we strongly discourage it; there can be issues that occur on device but not on the simulator and of course if you have a bug your app will be rejected.
That said, many people don’t have every device to test on and if you don’t have access to a NOOK but it’s working on all devices you’ve tested I’d think you’d probably be OK - still, if you can find a way to borrow a NOOK off a friend or the like it is always a good idea.
